These data provide systematic evidence on potency and quality of latanoprost ophthalmic solution LOS ; when subjected to various temperature and environmental conditions over time and were used to establish storage and transportation guidelines. METHODS The first study simulated conditions during manufacturing, transportation, and storage. LOS was cycled between 8, 25, and 40C temperatures for 36 months, then exposed for an additional 3 months to room temperature 25C ; and conditions simulating patient use. In two additional studies, LOS was subjected to continuous room temperature 25C ; conditions for 12 months and to freezing temperatures -20C ; for 6 months. Various chemical, physical, and microbiological parameters were tested for all stability evaluations, including assays for latanoprost, acid of latanoprost, and benzalkonium chloride. RESULTS In this 39-month study designed to evaluate the long-term effects of LOS storage at refrigerated, room, and elevated temperatures, and conditions simulating patient use, all parameters tested for quality and potency were within the manufacturing specifications for all sample groups evaluated. In the 12-month study of LOS exposure to continuous roomtemperature conditions at 25C, all parameters tested were within specification limits, including latanoprost concentrations range 92% to 103% of the labeled amount ; . Following exposure to -20C for 6 months, the assay for latanoprost remained within specification limits and there were no stabilityrelated changes in the parameters evaluated. CONCLUSIONS LOS was stable and retained its quality and potency when cycled between 8, 25, and 40C temperatures for 36 months. After 36 months, LOS was stable for an additional 3 months when exposed to room temperature 25C ; and conditions simulating patient use. Stability was also demonstrated when LOS was subjected to freezing temperatures -20C ; for 6 months, and to continuous room temperature 25C ; exposure for 12 months.
